Product Specific Information

The eBio17B7 antibody reacts with mouse and rat IL-17A, showing no cross-reactivity with IL-17F. IL-17A is a CD4+ T cell-derived cytokine that promotes inflammatory responses and is elevated in autoimmune and inflammatory conditions such as rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested by intracellular staining and flow cytometric analysis of restimulated, Th17-polarized splenocytes using the intracellular Fixation and Permeabilization Buffer Set (Product # 88-8824-00).
• Recommended usage ≤ 0.03 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
• Optimal titration recommended for best performance.

Staining can also be performed using the Foxp3 buffer system (Product # 00-5523-00).

Light Sensitivity:

• Tandem dye sensitive to photo-induced oxidation. Protect from light.

Fixation:

• Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark.

Target Information

Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine promoting inflammatory responses and is elevated in autoimmune diseases. IL-17A is a 32 kDa disulfide-linked homodimer of 136 amino acids, signaling through IL-17R/CDw217. It induces IL-6, IL-8, G-CSF, and PGE2 secretion in various cell types and supports neutrophil maturation. IL-17A regulates NF-κB and MAPK activities, enhances COX-2 and nitric oxide production.
