Target Information
Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are common in excitable cells and mediate communication between surface and intracellular ion channels.
Junctophilins (JPs) are essential components of these complexes. They consist of a C-terminal hydrophobic segment spanning the ER/SR membrane and a cytoplasmic domain with affinity for the PM.
Four JP subtypes are known:
- JPH1: Expressed in skeletal muscle
- JPH2: Found in all muscle cell types
- JPH3, JPH4: Predominantly expressed in the brain
In the CNS, JPH3 and JPH4 contribute to subsurface cistern formation in neurons. Mice lacking both JPH3 and JPH4 show impaired learning, memory, and abnormal neural function. A repeat expansion in JPH3 is associated with Huntington disease-like 2. At least two isoforms of JPH3 exist.
