Target Information

Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase subfamily that includes EGFR, ERBB2, and ERBB3.
ERBB4 binds and is activated by neuregulins and other factors, inducing various cellular responses such as mitogenesis and differentiation.
It is a receptor for heregulin and mediates tyrosine phosphorylation upon activation. Like other protein kinases, ERBB4 regulates cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.

ERBB4 is predominantly expressed in several breast carcinoma cell lines and in normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum.
High mRNA levels are found in T47-D, MDA-MB-453, BT-474, and H3396 cell lines, while intermediate levels occur in MCF-7, MDA-MB-330, and MDA-MB-361.
Low or absent expression is observed in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3.
Elevated ERBB4 levels in certain tumor cell lines suggest a potential role in malignancies.
