Product Specific Information
- In Western Blot, a band is seen at ~56 kDa representing the ATG5–ATG12 complex. The molecular weight of human ATG5 is ~33 kDa.
- For paraffin-embedded tissues, antigen retrieval using sodium citrate buffer (pH 6.0) is recommended.
- Suggested positive controls: antigen standard for ATG5 (transient overexpression lysate), mouse wildtype ES cell lysate.
Target Information
ATG5 (Autophagy Related 5) is a key component in the autophagy pathway and plays an important role in apoptosis. It is involved in mitochondrial quality control, regulation of innate antiviral immune responses, lymphocyte development, MHC II antigen presentation, and adipocyte differentiation.
Following conjugation to ATG12, the ATG5–ATG12 complex participates in autophagosome formation and contributes to autophagic cell death via interaction with FADD. It also associates with innate immune response proteins such as RIG-I and VISA (IPS-1), modulating type I interferon production. Dysregulation of ATG5 is linked to diseases such as spinocerebellar ataxia.
