Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. Upon stress signals, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

The p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells, acting as an anticancer agent. It is also linked to aging, oxidative stress, and memory-related processes (LTP).

In normal cells, p53 levels are low, but in transformed cell lines, expression is high and may contribute to malignancy. Mutations in p53 often result in loss of DNA-binding ability and tumor suppressor function. Alterations in the TP53 gene occur as somatic mutations in cancers and as germline mutations in syndromes such as Li-Fraumeni.
