Target Information
STAT3 (Signal Transducer and Activator of Transcription 3) is a member of the STAT protein family. It is phosphorylated by receptor-associated kinases and translocates to the nucleus, acting as a transcription factor in response to cytokines and growth factors. Coactivators such as CREB-binding protein are required for transcriptional activation by STAT3.
STAT3 can be activated by Interferon-alpha, Interferon-gamma, EGF, PDGF, and IL6. Phosphorylation on tyrosine 705 by JAK1 and JAK2 is essential for dimer formation, nuclear translocation, and DNA binding. The STAT3 gene is located on chromosome 17q in humans. It mediates expression of various genes involved in cell growth and apoptosis. Rac1 and PIAS3 regulate STAT3 activity, and three alternatively spliced transcript variants encoding distinct isoforms have been described. Deletion of the STAT3 gene in mice is lethal at the early embryonic stage.
