Mutations, rearrangements, and amplifications in the ALK gene are associated with various tumors, including:

- Neuroblastoma
- Non-small cell lung cancer
Chromosomal rearrangement between ALK (chromosome 2) and NPM (chromosome 5) results in the NPM-ALK fusion gene, which has oncogenic properties.
ALK exhibits structural similarity to LTK and functions as a receptor protein-tyrosine kinase with transmembrane and extracellular domains, essential for neuronal development.
