• Specificity Comments:
  Recognizes the epsilon-chain of CD3, composed of five polypeptide chains (γ, δ, ε, ζ, η; MW 16–28 kDa). The CD3 complex associates with the T cell antigen receptor (TCR) and is involved in signal transduction following antigen recognition.
  CD3 antigen appears early in thymocyte development, marking T cell lineage commitment. In cortical thymocytes, CD3 is mainly intracellular, while in medullary thymocytes, it appears on the surface.
  CD3 is a highly specific T-cell marker, present in most T-cell neoplasms.
  Does not react with Cat or Dog.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T-cell cytoplasm and regulates TCR complex expression.
T-cell activation via TCR involves CD3γ, CD3δ, CD3ε, and CD3ζ subunits, members of the immunoglobulin superfamily encoded on chromosome 11.
Their cytoplasmic tails associate with signaling molecules through a double tyrosine-based motif.
CD3 may play roles in TCR-induced growth arrest, survival, and proliferation.
CD3 antigen is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells.
Decreased T lymphocytes may be observed in autoimmune diseases.
Defects in CD3 genes are associated with CD3 immunodeficiency.
