Target Information
PAK 단백질은 Rho GTPase를 세포골격 재조직화 및 핵 신호전달과 연결하는 주요 효과기입니다. PAK 단백질은 세린/트레오닌 p21-활성화 키나아제 패밀리에 속하며, 소형 GTP 결합 단백질 Cdc42와 RAC의 표적 역할을 합니다. α, β, γPAK 이소폼은 활성화된 GTP 결합 상태의 Rac1 및 Cdc42와 특이적으로 복합체를 형성하며, 이들의 GTPase 활성을 억제하고 자가 인산화를 유도합니다. γPAK에는 Ser19, Ser141, Thr402를 포함한 8개의 자가 인산화 부위가 있으며, Ser141 및 Thr402의 인산화는 γPAK 활성화와 관련이 있습니다. 인산화 후 Rac/Cdc42에 대한 친화도가 감소하면, PAK 이소폼은 복합체에서 분리되어 하위 기질로 이동합니다.
