Target Information

Cas9 is an RNA-guided DNA endonuclease enzyme associated with the CRISPR adaptive immunity system in bacteria and in genetic engineering.
The system uses RNA to direct the nuclease to a specific DNA sequence, enabling indel mutations, sequence insertions, deletions, or rearrangements.
Cas9 can also mediate gene upregulation or epigenetic modifications.

Streptococcus pyogenes Cas9 (SpCas9) and Staphylococcus aureus Cas9 (SaCas9) are common variants.
SaCas9 is smaller (1053 AA vs. 1368 AA), efficient, and specific, recognizing an NNGRRT PAM and producing comparable indel rates to SpCas9.
This makes SaCas9 advantageous for all-in-one delivery with adeno-associated vectors.
