Target Information
CYLD는 세포질 내 탈유비퀴틴화 효소로, ubiquitin carboxy-terminal hydrolases (UCH) 계열에 속하며 세 개의 CAP-GLY 도메인, proline-rich 영역, SH3 결합 도메인 및 UCH 촉매 도메인과의 서열 상동성을 가진 956 아미노산 단백질입니다.
CYLD는 종양 억제 단백질로서 JNK 신호 전달 경로에 영향을 미치며, TRAF2 및 NF-kappa-B 신호 경로의 음성 조절자로 작용합니다. 또한 I-kappa-B kinase 경로의 수용체 의존적 조절 역할을 가지고 있으며, Lys-48이 아닌 polyubiquitin 사슬에 대해 탈유비퀴틴화 활성을 나타냅니다. TRAP1은 CYLD의 새로운 탈유비퀴틴화 기질로 알려져 있습니다.
CYLD 변이는 cylindromatosis, multiple familial trichoepithelioma, Brooke-Spiegler syndrome과 관련이 있습니다.
