Target Information
Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene. It is a key component of the extracellular matrix in cartilaginous tissue, providing resistance to compression.
Human Aggrecan is approximately 250 kDa (2316 amino acids) and exists in multiple isoforms via alternative splicing. It has a bottlebrush structure with chondroitin sulfate and keratan sulfate chains attached to the core protein.
Aggrecan synthesis and degradation are closely linked to cartilage deterioration during joint injury, disease, and aging. The protein contains three interglobular domains (G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.
Aggrecan fragments are released into synovial fluid during osteoarthritis due to cleavage by aggrecanase and MMP activity. Antibodies BC-3 and BC-4 detect neo epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
