Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA.

When bound to EREs, ER can regulate gene transcription positively or negatively through recruitment of coactivator or corepressor proteins. Two forms exist—alpha and beta—encoded by ESR1 and ESR2 genes, respectively. Alternative RNA splicing produces at least four known estrogen receptor-alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa).

Estrogen receptors are widely expressed in various tissues, essential for sexual development and reproductive function, and also play roles in bone and other tissues. They are implicated in pathological processes including breast cancer, endometrial cancer, and osteoporosis.
