Target Information
The PAX gene family encodes transcription factors active during early embryonic development. Members possess a DNA-binding domain encoded by the paired box and regulate genetic programs essential for tissue differentiation.
Several PAX genes are linked to developmental disorders:
- PAX3: associated with Waardenburg syndrome
- PAX6: associated with Aniridia
- PAX2: associated with Wilms tumor
During embryogenesis, PAX2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Expression decreases as the tubular epithelium matures. Persistent expression of PAX2 in Wilms tumors correlates with poorly differentiated epithelial cell proliferation. WT1, the Wilms tumor suppressor protein, is thought to repress PAX2 transcription.
