Target Information
ZAP-70 is a cytosolic protein tyrosine kinase in the Syk family, expressed in T-cells and natural killer cells. It is essential for T-cell receptor activation.
Upon TCR engagement, ZAP70 is phosphorylated on tyrosines 292, 315, and 319 in the interdomain B region.
- Tyr315 (Vav binding site) and Tyr319 (Lck binding site) enhance ZAP70-mediated lymphocyte signaling.
- Tyr292 phosphorylation attenuates activation, regulating signaling duration.
