Target Information
Tau는 주로 축삭에 존재하는 신경세포 미세소관 결합 단백질로, 튜불린 중합을 촉진하고 미세소관을 안정화하는 역할을 합니다.
C-말단은 축삭 미세소관에, N-말단은 신경 세포막 성분에 결합하여 두 구조를 연결하는 역할을 합니다.
TAU/MAPT의 세포 내 위치는 축삭 극성을 결정하며, 짧은 isoform은 세포골격의 유연성을, 긴 isoform은 안정성을 담당합니다.
과인산화된 Tau는 알츠하이머병(AD) 뇌의 신경섬유 엉킴(neurofibrillary tangles)을 구성하는 주요 성분으로, 미세소관 결합 기능을 손상시켜 신경세포 퇴화를 유발합니다.
Tau의 인산화에는 GSK-3β, PKA, cdk5, casein kinase II 등의 효소가 관여하며, Pick병, 전두측두치매, 피질기저변성, 진행성핵상마비 등 다양한 신경계 질환에서도 발견됩니다.
