Target Information
Machado-Joseph disease (MJD) is an autosomal dominant neurologic disorder, also known as spinocerebellar ataxia-3. The MJD protein (Ataxin-3) contains CAG repeat expansions in the coding region, where expansion from normal 13–36 repeats to 68–79 causes the disease. There is a negative correlation between age of onset and CAG repeat number. Ataxin-3 interacts with transcription regulators (CBP, p300, PCAF), represses transcription, and functions as a histone-binding and deubiquitinating enzyme.
