Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Immunoglobulin classes share a basic four-polypeptide chain structure consisting of two heavy chains and two light chains (kappa, lambda; both ~22.5 kDa).
Kappa and lambda chains have variable and constant regions, distinguishable by the antigenic properties of the constant region.
The typical kappa-to-lambda ratio is 70:30.
Antibodies to the kappa light chain are useful for identifying leukemias, plasmacytomas, and certain non-Hodgkin’s lymphomas.
Demonstration of clonality in lymphoid infiltrates indicates malignancy.
