Target Information
STAT3는 STAT(signal transducers and activators of transcription) 단백질 계열에 속하며, receptor-associated kinase에 의해 인산화되어 핵으로 이동 후 cytokine 및 성장인자 자극에 반응하여 전사 인자로 작용합니다. STAT3의 전사 활성화에는 CREB-binding protein 등의 공활성화 인자가 필요하며, Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있습니다. Tyrosine 705의 인산화는 JAK1, JAK2에 의해 매개되며, STAT3의 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다.
인간에서는 STAT3 유전자가 17번 염색체 q arm에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다. STAT3는 c-Jun 및 CREB와 상호작용하며, 세포 성장과 아폽토시스 등 다양한 세포 과정에 관여합니다. 또한 Rac1 GTPase가 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제 인자입니다. STAT3는 세 가지 대체 스플라이싱 변이체를 가지며, knockout 마우스에서는 배아 발생 초기 단계에서 치명적입니다.
