Target Information

Perforin is a major cytolytic protein found in cytolytic granules and plays a critical role in immune defense against tumors and viral infections. It is a 555 amino acid protein (70–75 kDa) with a 21 amino acid signal peptide. Perforin forms transmembrane pores in target cells, similar to complement component C9. It is expressed in cytotoxic T lymphocytes and natural killer cells, but not in helper T cells or other tumor cell types. Mutations in the perforin gene are associated with familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder in early childhood. Alternative splicing results in multiple transcript variants.
