Target Information
Cas9 is an RNA-guided DNA endonuclease enzyme associated with the CRISPR adaptive immunity system in bacteria and genetic engineering.
This system uses RNA to guide the nuclease to a desired DNA sequence, allowing for targeted indel mutations, sequence insertions, deletions, or rearrangements.
SaCas9, derived from Staphylococcus aureus, offers several advantages for genome editing:
- Small size and high efficiency
- Nickase activity and high specificity
- Recognizes NNGRRT PAM sequence
- Comparable cleavage rate to SpCas9 with spacer lengths of 20–24 nucleotides
Compared with SpCas9, SaCas9 (1053 AA) is smaller than SpCas9 (1368 AA) and provides efficient indel generation.
It is preferred for all-in-one delivery systems using adeno-associated vectors for Cas9 and multiple gRNA expression cassettes.
