Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule. CD11b associates with integrin beta2 (CD18) and is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It functions as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, playing a crucial role in cell-cell and cell-matrix interactions.

Each product contains:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.4 µg) per 100 µL test volume. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD11b (integrin alpha-M, ITGAM) forms a heterodimer with CD18 (integrin beta-2), known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cell subsets, dendritic cells, and microglia.
CD11b/CD18 acts as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating essential adhesion processes.
Expression levels vary by cell type and maturation stage. It is a common microglial marker and is implicated in systemic lupus erythematosus and related immune disorders.
