Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule. CD11b, associated with integrin beta2 (CD18), is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It serves as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, playing a key role in cell-cell and cell-matrix interactions.

Each product includes:

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use 5 µL (0.6 µg) per test in a final 100 µL volume. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing engineered nucleic acid dye structures to minimize spectral spillover and spread.

Target Information

CD11b (integrin alpha-M, ITGAM) is a 165 kDa adhesion molecule that forms a heterodimer with integrin beta-2 (CD18), known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. CD11b/CD18 binds ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating critical adhesion and immune interactions.

Expression varies by tissue:

• 8% of spleen cells
• 44% of bone marrow cells
• <1% of thymocytes
  Commonly used as a microglial marker in nervous tissue.
  CD11b expression increases during monocyte maturation, with higher levels in peritoneal macrophages than splenic macrophages.
  Associated diseases include systemic lupus erythematosus and ITGAM-related autoimmune susceptibility.
