Description:
This monoclonal D9302C12 antibody reacts with and inhibits the bioactivity of mouse IL-9. IL-9 is a 14 kDa cytokine originally named P40, identified by its proliferative effects on T cell populations. The IL-9 receptor, a heterodimer of the gamma chain of the IL-2 receptor and the IL-9R chain, activates Jak/STAT signaling pathways upon ligand binding. IL-9 induces Th17 and Treg differentiation in CD4+ T cells, IgE production in B cells, and mast cell proliferation. IL-9 expression occurs in Th2, Th17, eosinophil, and mast cells. Th9 cells, a subset of CD4+ T cells, secrete large amounts of IL-9 and IL-10, and their development is induced by IL-4 and TGF-β stimulation.

The D9302C12 antibody inhibits the bioactivity of mouse IL-9.
Applications Tested:
The ND50 of D9302C12, measured by inhibition of MC/9 cell proliferation, is ≤500 ng/mL in the presence of 0.1 ng/mL of Mouse IL-9 Recombinant Protein. Neutralization dose varies depending on assay method, cytokine concentration, and cell type. Titrate carefully for optimal performance.

Target Information
Interleukin 9 (IL-9) is produced by activated T helper 2 (Th2) lymphocytes and regulates multiple cell types involved in Th2-associated asthma responses, including B and T lymphocytes, mast cells, eosinophils, and epithelial cells. Mouse IL-9 cDNA encodes a 144 amino acid precursor with an 18 amino acid signal peptide, forming a mature 14 kDa cysteine-rich protein. Native mouse IL-9 is heavily glycosylated. TGF-β plays an important role in the differentiation of IL-17-producing T helper cells and inducible regulatory T cells by reprogramming Th2 cells to secrete IL-9, promoting Th9 cell differentiation.
