Target Information
HMHA1 단백질은 Rho-type GTPases의 GTPase activator (RhoGAP) 도메인을 포함하여 actin cytoskeleton 및 세포 확산을 음성 조절할 수 있습니다. 또한 N-말단 BAR 도메인을 포함하여 RhoGAP 활성을 자가 억제할 수 있습니다.
이 단백질은 minor histocompatibility antigen HA-1의 전구체로, mHag은 MHC와 결합 시 특정 T-cell에 의해 인식되어 면역 반응을 유도하는 면역원성 펩타이드입니다. 이러한 펩타이드는 항원 처리 경로를 통해 절단되어 MHC class I 또는 II 분자에 결합 후 세포 표면에 발현되어 T-cell 반응을 자극하고, 골수 이식 후 이식편 거부반응(GVHD)을 유발할 수 있습니다.
특히 HA-1 항원의 불일치는 HLA-identical BMT 후 심각한 GVHD 발생과 관련이 있으며, HA-1은 MHC class I HLA-A*0201 및 다른 HLA-A allele에 의해 세포 표면에 제시됩니다. 이 복합체는 공여자 CTL 반응을 유발하여 조혈계 악성종양에 대한 세포 인식 및 용해를 일으킵니다.
