Target Information

HMHA1 contains a GTPase activator for the Rho-type GTPases (RhoGAP) domain, which can negatively regulate the actin cytoskeleton and cell spreading. It also contains an N-terminal BAR-domain that exerts an autoinhibitory effect on RhoGAP activity.
This protein is a precursor of the histocompatibility antigen HA-1. Minor histocompatibility antigens (mHags) are immunogenic peptides that, when complexed with MHC, can elicit immune responses recognized by specific T-cells. These peptides, derived from polymorphic intracellular proteins, are processed and presented by MHC class I or II molecules, potentially triggering graft rejection or graft-versus-host disease (GVHD) after hematopoietic stem cell transplantation from HLA-identical donors.

Mismatching for mHag HA-1, recognized as immunodominant, is associated with severe GVHD after HLA-identical bone marrow transplantation. HA-1 is presented by MHC class I HLA-A*0201 and other HLA-A alleles, eliciting donor cytotoxic T-lymphocyte (CTL) reactivity against hematologic malignancies following allogenic BMT, leading to specific cell recognition and lysis by CTL.
