Description:
This 1H7 monoclonal antibody reacts with human insulin-like growth factor-1 receptor (IGF-1R), also known as CD221. This receptor is a cell surface-expressed glycoprotein composed of two extracellular alpha subunits and two transmembrane beta subunits with tyrosine kinase activity. Expressed on nearly all cell types, IGF-1R binds IGF-I, IGF-II, and insulin, activating PI3K/Akt and MAPK pathways involved in cell proliferation and survival. Many tumors and transformed cells display altered IGF-1R expression.

This antibody has been reported to block IGF-I and IGF-II binding to IGF-1R.

Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of the HeLa cell line. Recommended usage is 5 µL (0.25 µg) per test. A test is defined as the amount of antibody required to stain a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Target Information

IGF1R (Insulin-like Growth Factor 1 Receptor):
A receptor tyrosine kinase activated by IGF-1 and IGF-2, implicated in various cancers. The protein is a tetramer consisting of two alpha and two beta subunits. The alpha subunits bind IGF-1 with higher affinity than IGF-2. Binding induces conformational change and autophosphorylation at residues 1134–1136, leading to phosphorylation of substrate proteins and activation of RAS, RAF, and MAPK pathways (ERK, p38, JNK), promoting gene transcription for proliferation.
