The coronavirus Spike (S) protein is crucial for viral attachment, fusion, and entry, making it a key target for antibody, inhibitor, and vaccine development.
The receptor-binding domain (RBD, S-RBD) of the Spike protein binds strongly to angiotensin-converting enzyme 2 (ACE2) receptors in humans and bats.
SARS-CoV-2 RBD shows significantly higher binding affinity to ACE2 compared to SARS-CoV RBD.
The S1 subunit contains the RBD that specifically interacts with ACE2 on target cells.
