Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family.
It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and participate in receptor internalization via ubiquitin-mediated mechanisms.
- Expressed on cells of myelomonocytic lineage
- Binds sialic acid residues in N- and O-glycans
- Therapeutic target for acute myeloid leukemia
- Found on granulocyte and macrophage precursors in bone marrow
- Absent on pluripotent stem cells
- Useful marker for peripheral monocytes
- Helps distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
