Target Information
The low density lipoprotein receptor (LDL-R) gene family consists of cell surface proteins involved in receptor-mediated endocytosis of specific ligands. LDL binds at the cell membrane and is internalized, leading to degradation in lysosomes and release of cholesterol. This process regulates HMG CoA reductase activity, the rate-limiting step in cholesterol synthesis, and stimulates cholesterol ester formation. Mutations in LDL-R cause familial hypercholesterolemia. LDL-R also acts as an entry factor for Hepatitis C virus along with SCARB1, CLDN1, and CD81. At least three isoforms of LDL-R are known.
