Target Information
The BBSome complex functions as a coat complex required for sorting specific membrane proteins to the primary cilia. It is essential for ciliogenesis but not for centriolar satellite function. This function involves the Rab8 GDP/GTP exchange factor, which localizes to the basal body and interacts with the BBSome. Rab8(GTP) enters the primary cilium, promoting ciliary membrane extension. The BBSome associates with the ciliary membrane, binds to RAB3IP/Rabin8 (the GEF for Rab8), and facilitates docking and fusion of carrier vesicles to the ciliary membrane base. Together with LTZL1, the BBSome regulates SMO ciliary trafficking and contributes to the Sonic Hedgehog (SHH) pathway regulation. It is required for BBSome complex ciliary localization but not for proper complex assembly.
