Target Information

Furin is a membrane-associated, calcium-dependent serine protease belonging to the subtilisin-like prohormone convertase (PC) family. These enzymes cleave prohormones and neuropeptide precursors, as well as various cellular, viral, and bacterial proteins transported via the constitutive secretory pathway.

Furin and other PC family members share structural similarities, including:

• A heterogeneous ~10 kDa amino-terminal proregion
• A conserved ~55 kDa subtilisin-like catalytic domain
• A variable carboxyl-terminal domain

Furin becomes catalytically active after proregion cleavage in the appropriate cellular compartment and is the only PC known to possess a transmembrane domain. Cleavage of target proteins occurs at the carboxyl-terminus of the furin consensus sequence RX(K/R)R. The peptide sequence C771PSDSEEDEG780 localizes furin to the trans-Golgi network, and phosphorylation of serine residues in this region modulates intracellular routing. The tetrapeptide Y759KGL762 directs internalization from the cell surface.
