Target Information
Furin은 막 결합형 칼슘 의존성 세린 프로테아제로, subtilisin-like prohormone convertase (PC) 계열에 속합니다. 이 효소들은 대부분의 프로호르몬과 뉴로펩타이드 전구체를 절단하며, 다양한 세포 및 바이러스 단백질, 세균 독소의 성숙에도 관여합니다.
Furin은 약 10 kDa의 N-말단 proregion, 약 55 kDa의 subtilisin-like catalytic domain, 그리고 길이와 서열이 다양한 C-말단 도메인을 포함합니다. Proregion 절단 후 활성화되며, PC 중 유일하게 transmembrane domain을 갖습니다.
Furin은 RX(K/R)R 서열의 C-말단에서 기질 단백질을 절단하며, C771PSDSEEDEG780 서열은 trans-Golgi-network로의 위치를 결정합니다. 또한 Y759KGL762 서열은 세포 표면으로부터의 내재화를 유도합니다.
