Product Specific Information

This antibody recognizes an extracellular/luminal epitope of CD107a, an approximately 100–120 kDa glycoprotein expressed mainly on lysosomal membranes and also on the plasma membrane.

Target Information

LAMP1 (CD107a, lysosome-associated membrane protein-1) is a major constituent of lysosomal membranes along with LAMP2. It is a heavily glycosylated type I integral membrane protein containing:

  • A putative signal peptide
  • 18 N-linked glycosylation sites
  • A single transmembrane domain
  • A short cytosolic tail (11 amino acids)

LAMP1 plays a role in lysosome biogenesis and fusion with phagosomes. Upon cell activation, LAMP1 translocates to the plasma membrane via a carboxyl-terminal tyrosine-based motif (YXXI). Mutations affecting this motif disrupt lysosomal localization and cause cycling between plasma membrane and endosome.

Cell surface LAMP1 and LAMP2 promote adhesion of human PBMCs to vascular endothelium, suggesting involvement in inflammation processes. Increased LAMP1 expression is observed in neurons and glial cells near senile plaques in Alzheimer’s Disease, and in medullary epithelial cells, macrophages, and lymphocytes during acute thymic involution. LAMP1 serves as a reliable marker of mast cell activation.
