Target Information
CD35 (CR1, Complement Receptor 1) is a cell membrane-bound, monomeric glycoprotein that serves as the receptor for C3b and C4b, key components of the complement system. It mediates binding to particles and immune complexes that have activated complement.
The CD35 complex has four different allotypes (C, A, B, D):
- C: 160 kDa
- A: 190 kDa
- B: 220 kDa
- D: 250 kDa
CD35 is involved in immune complex processing, promoting binding and phagocytosis of C3b complexes, and inhibiting complement activation. It is expressed on neutrophils, eosinophils, monocytes, B cells, some NK cells, erythrocytes, myeloid leukemias, and follicular dendritic reticulum cells, but not on basophils.
Reduced expression or mutations in CD35 have been associated with gallbladder carcinoma, mesangiocapillary glomerulonephritis, systemic lupus erythematosus, and sarcoidosis. Mutations also correlate with reduced Plasmodium falciparum rosetting, conferring protection against severe malaria. Alternate allele-specific splice variants and a secreted form have been described.
