Target Information
Arginase-1 (Arg1) is a 35 kDa enzyme that converts L-arginine to urea and L-ornithine, the final step in the urea cycle. The resulting polyamines are essential for cell proliferation and detoxification of protein degradation byproducts. By degrading arginine, Arginase 1 reduces nitric oxide production by depriving NO synthase of its substrate.
In humans and mice, Arginase 1 is expressed in the liver, neutrophils, myeloid-derived suppressor cells (MDSCs), and neural stem cells. In humans, it is found in blood neutrophils but not CCR3+ granulocytes. In mice, Arginase 1 is a hallmark of alternatively activated macrophages (M2a).
Arginase-1 may also be expressed in myeloid cells infiltrating tumors and is typically found in most hepatocellular carcinomas. Defects in Arginase 1 cause argininemia, an autosomal recessive disorder characterized by hyperammonemia.
