Target Information
ICAD (Inhibitor of Caspase-Activated DNase) is a substrate that regulates nuclear apoptosis. It has two isoforms:
- ICAD-L (DFF45, 45 kDa) – functional isoform
- ICAD-S (DFF35, 35 kDa) – shorter isoform
Both isoforms can bind and inhibit CAD (Caspase-Activated DNase), but only ICAD-L is functional. During apoptosis, ICAD is cleaved by caspase-3 at residues 117 and 224, releasing CAD to execute DNA fragmentation. In non-apoptotic cells, CAD remains complexed with ICAD. ICAD also functions as a folding chaperone specific for CAD. Recent research suggests ICAD as a potential therapeutic target for restoring apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
