Target Information
Caspase 8 (CASP8)는 cysteine-aspartic acid protease (caspase) 계열에 속하며, 세포 사멸(apoptosis)의 실행 단계에서 중심적인 역할을 합니다. Caspase는 비활성 전구체 형태로 존재하며, 내부의 aspartic residue에서 단백질분해 처리되어 활성화됩니다. Caspase 8은 Fas 및 다양한 세포사멸 자극에 의해 유도되는 programmed cell death에 관여하며, FADD 단백질과 상호작용하는 death effector domain(DED)을 포함합니다. 또한 Huntington 병 환자의 뇌 조직에서 비정상적으로 검출되어 신경퇴행성 질환과의 연관성이 시사되었습니다. 활성화된 Caspase 8은 Caspase 9을 비롯한 하위 caspase들을 활성화시켜 세포 사멸을 유도합니다. Jurkat 세포 및 편도(Tonsil)에서도 반응성이 있으며, 과발현 시 apoptosis를 유도합니다. ICE family 억제제에 의해 차단될 수 있습니다. 다양한 splice variant가 보고되었으나, 모든 isoform의 full-length 서열은 확인되지 않았습니다.
