Target Information
Tight junctions are specialized regions of cell-cell contact abundant in luminal epithelial sheets. In freeze-fracture electron micrographs, they appear as belt-like bands of sealing strands that encircle the lateral surfaces of cells. These strands interact to form a molecular gasket preventing ions, water, and molecules from leaking between cells.
Beyond the barrier function, tight junctions maintain epithelial polarity by blocking diffusion of membrane proteins between apical and basolateral surfaces. Several peripheral membrane proteins (ZO-1, ZO-2, ZO-3, cingulin, Rab-3b, symplekin) are involved in assembly, maintenance, and permeability regulation. Actin filaments also play a major role in regulating tight junction permeability.
Claudin-1 and Claudin-2 are key structural components of tight junction strands. Claudin-2 functions as a paracellular channel selective for cations (Na+), mainly expressed in liver and kidney, with minor expression in the brain.
