Target Information
PUMA는 Bax 및 Noxa와 함께 p53의 전사적 표적에 속하는 친사멸성 Bcl-2 패밀리 단백질 중 하나입니다. PUMA 유전자는 BH3 도메인을 포함하는 두 단백질, PUMA-a와 PUMA-b를 암호화합니다. 이 단백질들은 Bcl-2에 결합하고 미토콘드리아에 위치하며, p53 반응 시 시토크롬 c 방출과 세포사멸을 유도합니다. PUMA는 p53 유도 세포사멸의 직접적인 매개체로 작용할 수 있습니다. PUMA 발현의 antisense 억제는 p53에 대한 세포사멸 반응을 감소시키며, 이는 PUMA가 시토크롬 c/Apaf-1 의존 경로를 통한 p53 유도 세포사멸에 관여함을 시사합니다.
