Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich (SRCR) superfamily. It functions as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and participating in iron recycling via endocytosis and heme breakdown.
CD163 is expressed on the surface of human monocytes and macrophages, especially during the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, excluding mantle zone and germinal center macrophages, interdigitating reticulum cells, and Langerhans cells. Expression is higher on CD16⁺ monocytes and is upregulated by IL-10, IL-6, and dexamethasone. Conversely, lipopolysaccharide (LPS) and phorbol myristate acetate (PMA) induce shedding of CD163 into plasma or cell supernatant.
CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for complexes involving multimeric haptoglobin (HP1F) than for dimeric haptoglobin (HP1S). It also triggers intracellular signaling cascades involving tyrosine kinase–dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
