The monoclonal antibody 8B6 recognizes human Placental Alkaline Phosphatase (PLAP), a 130 kDa enzyme expressed by placental syncytiotrophoblastoid cells. This enzyme exhibits slightly different kinetics from other phosphatases and can be found both on the plasma membrane and in the cytoplasm. Expression occurs in several germ line cancers (ovarian, testicular), human embryonic stem cells, and bone marrow-derived mesenchymal stromal cells.
This antibody reacts with both Regan and Nagao isozymes.
Applications Reported: Suitable for IHC (frozen and paraffin-embedded tissues) and ELISA.
Applications Tested: Tested on FFPE human placenta with trypsin digestion. Recommended ≤20 µg/mL; titration advised for optimal performance.

eFluor® 660 replaces Alexa Fluor® 647. It emits at 659 nm and is excited with a red laser (633 nm). Ensure instrument compatibility.

Target Information
Placental Alkaline Phosphatase (PLAP) regulates specific inflammatory processes and exists as one of four related alkaline phosphatases: intestinal, placental, placental-like, and liver/bone/kidney forms. PLAP (Regan and Nagao type) is a membrane-bound isoenzyme expressed in the placenta during the third trimester. It aids in identifying testicular germ cell tumors. Unlike germ cell tumors, PLAP-positive somatic cell tumors express epithelial membrane antigen (EMA).
PLAP may play a role in matrix mineralization, though mice lacking this enzyme show normal skeletal development. Mutations in PLAP are linked to hypophosphatasia, a disorder characterized by hypercalcemia and skeletal defects, with variable severity based on mutation type.
