Product Specific Information
This HECA-452 monoclonal antibody reacts with human Cutaneous Lymphocyte Antigen (CLA), a cell surface glycoprotein predominantly expressed on T cells in the skin. CLA can also be detected on peripheral T and NK cells, monocytes, neutrophils, and CD1a+ dendritic cells. The antibody recognizes sialofucosylated glycans and sialyl Lewis x (sLex), which are involved in adhesion and infiltration of T lymphocytes at sites of cutaneous inflammation.
HECA-452 has been reported to inhibit sLex binding to E-selectin, though this remains debated.
Applications include flow cytometry, western blotting, and immunohistochemical staining of FFPE tissue sections. Tested by flow cytometry on normal human peripheral blood cells and by IHC on FFPE human tissue at ≤20 µg/mL. Optimal performance with low pH antigen retrieval; high pH retrieval also applicable.
eFluor™ 660 replaces Alexa Fluor™ 647, emitting at 659 nm and excited by the red laser (633 nm).

Target Information
Cutaneous Lymphocyte Antigen (CLA) is mainly expressed on skin T cells but can also be found on peripheral T and NK cells, monocytes, neutrophils, and CD1a+ dendritic cells. It recognizes sialofucosylated glycans and sialyl Lewis x (sLex), binding to E-selectin and ELAM-1, mediating lymphocyte adhesion and infiltration in skin inflammation.
