Product Specific Information

The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.
In B cells, IRF4 is highly expressed in mature plasma cells and regulates differentiation through interaction with PU.1 and activation of genes such as Prdm1 (Blimp1).
In T cells, IRF4 contributes to Treg, Th2, Th9, and Th17 cell development and function, being upregulated upon activation. It induces Blimp1 and IL-10 expression in Tregs.
IRF4 also plays a role in macrophage polarization and immune regulation, and its overexpression has been linked to several hematologic malignancies.

Applications Tested: Intracellular staining and flow cytometry of stimulated human peripheral blood cells using the Foxp3/Transcription Factor Buffer Set.

Target Information

The interferon regulatory factor (IRF) family contains transcription factors with a conserved tryptophan pentad repeat DNA-binding domain.
IRFs regulate interferon expression in response to viral infection and control interferon-inducible gene expression.
IRF4 is lymphocyte-specific and negatively regulates toll-like receptor (TLR) signaling, a key pathway in innate and adaptive immunity.
A chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH loci is associated with multiple myeloma.
Alternative splicing variants have been identified.
