Product Specific Information
- Antibody is tested in direct ELISA.
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature (RT).
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at RT for 20 minutes.
Target Information
Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type 1 glycoprotein (40–43 kDa) expressed in many tumors and normal cells, particularly lymphatic endothelial and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissues and thymic epithelial cells.
Podoplanin regulates lymphatic endothelium structure and may maintain podocyte morphology. It acts as a ligand for CLEC-2 and is associated with tumor invasiveness and metastasis. The protein is implicated in cell migration, metastasis formation, and tumor invasion.
Podoplanin expression is induced via Src activation of Cas and has been detected on Th17 cells and a distinct subset of CD4+ PBMCs. Dysfunction is linked to lymphangioma and subglottis neoplasm, and it may serve as a marker for lung injury due to its physiological role.
