1 µg/mL of MA5-27549 was sufficient for detection of HDEL-containing proteins in 10 µg of S. cerevisiae lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody. Detects approximately 78 kDa.

Target Information

HSP 70 family comprises four highly conserved proteins: HSP 70, HSC 70, GRP 75, and GRP 78. These proteins act as molecular chaperones, assisting in multi-protein complex assembly, translocation across membranes, and proper folding of nascent chains.
GRP 78 is localized in the endoplasmic reticulum (ER), where it aids in folding and translocation of secretory proteins. Sorting of these proteins depends on a C-terminal tetrapeptide signal—KDEL in animal cells and HDEL in S. cerevisiae.
Clone 2E7 recognizes the C-terminal peptide HDEL, a common ER retention signal found in yeast, plants, nematodes, and other eukaryotic proteins. It specifically stains HDEL proteins in barnyard grass, beet, cotton, mung bean, sorghum, and wheat.
