Product Specific Information

The monoclonal antibody 87G recognizes human HLA-G, a member of the Human Leukocyte Antigen family involved in inhibiting immune responses. HLA-G has seven reported isoforms, and the 87G antibody recognizes both HLA-G1 and soluble HLA-G5.

HLA-G expression is primarily found in fetal trophoblast cells invading the maternal decidua, protecting the fetus from maternal immune attack. High expression has also been identified in certain tumors such as melanoma, breast carcinoma, renal carcinoma, CLL, AML, and B-CLL. Additional expression is observed in pancreatic islets, erythroid and endothelial progenitors, and the adult thymic medulla.

HLA-G⁺ CD4 or CD8 cells in normal human peripheral blood are thought to act as regulatory cells with a unique cytokine profile distinct from Tregs. The receptors for HLA-G include CD85j/ILT2, CD85d/ILT4, and CD158. Recent studies indicate a role for HLA-G in transplant tolerance and maintenance.

Target Information

HLA-G belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (β2-microglobulin). The heavy chain (~45 kDa) is membrane-anchored and encoded by a gene with 8 exons:

  • Exon 1: Leader peptide
  • Exons 2 & 3: α1 and α2 domains (peptide-binding)
  • Exon 4: α3 domain
  • Exon 5: Transmembrane region
  • Exon 6: Cytoplasmic tail

HLA-G is expressed on fetal-derived placental cells and plays a key role in immune tolerance. HLA and MHC antibodies are important tools in Immunopeptidomics, aiding neoantigen identification and characterization via LC-MS/MS.
