The A4 monoclonal antibody reacts with the class I human leukocyte antigen (HLA) complex in the absence of beta2 microglobulin.
There are three class I alpha-chain genes in humans, called HLA-A, -B, and -C. Normally, HLA class I proteins are associated non-covalently with beta2 microglobulin (β2M).
The function of the HLA class I complex is to present peptides derived from intracellular pathogens, including viral pathogens, on the cell surface to antigen-specific CD8+ cytotoxic T cells.
Class I HLA is expressed by the majority of nucleated cells.
Applications Reported: This A4 antibody has been reported for use in flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of stimulated normal human peripheral blood cells.
Recommended usage: 5 µL (0.125 µg) per test.
A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL.
Cell number should be determined empirically (10⁵–10⁸ cells/test).
