Target Information
TAK1 (also known as MAP3K7)는 serine/threonine kinase 계열에 속하는 다기능 단백질로, 다양한 신호 전달 경로에서 중요한 역할을 합니다.
TGF-β receptor 및 TRAF6과 상호작용하여 JNK 및 p38의 활성화를 매개하며, Wnt, BMP, activin 신호뿐 아니라 thyroid hormone, osmotic stress, endothelin, ephrine 반응에도 관여합니다.
TAK1은 von Hippel-Lindau tumor suppressor의 발현을 활성화하여 PDGF-B, integrin β1 및 β5를 억제함으로써 상처 치유를 조절합니다.
또한 IL-1, IL-6, IL-18, TNF, CD40, TLR, RIG-I 등 다양한 경로에서 IKK 활성화에 필수적이며, TAB2/3 및 ubiquitination을 통해 TAK1-IKK 경로가 활성화됩니다.
TAK1은 Thr178, Thr184, Thr187, Ser192에서 자가 인산화되며, IKK-β를 인산화합니다.
추가적으로, TAK1은 인간 telomerase reverse transcriptase를 억제하여 세포 수명 조절에 관여합니다.
