The S4B6 monoclonal antibody reacts with mouse interleukin-2 (IL-2), a 17 kDa factor mainly produced by activated CD4+ T cells. IL-2 induces antigen-nonspecific cell cycle progression and clonal expansion of activated T cells. It also acts on activated B cells, monocytes, NK cells, LAK cells, and oligodendroglial cells in vitro. IL-2 plays a central role in hematopoiesis, tumor surveillance, and anti-inflammatory responses.

The S4B6 antibody exhibits neutralizing activity and binds a different IL-2 epitope than JES6-1A12. In mice, S4B6 prevents IL-2 from binding to the high-affinity IL-2R but allows interaction with the low-affinity receptor. Conversely, IL-2 bound to JES6-1A12 can still interact with the high-affinity IL-2R.

Interleukin 2 (IL-2) is an immunomodulatory cytokine essential for proliferation of activated T cells, differentiation of B cells, NK cells, monocytes, and macrophages. IL-2 signals through the IL-2 receptor (IL-2R), a heterotrimeric complex whose gamma chain is shared with IL-4 and IL-7. Expression of the IL-2 gene in mature thymocytes is monoallelic, representing a unique regulatory mode for precise gene expression. Disruption of IL-2-like genes in mice leads to ulcerative colitis-like disease, highlighting its key role in immune response to antigens.
